Superpolymerase: development and application of a novel method for isothermal DNA amplification

In this project we aim to develop a new generation of DNA polymerase (“superpolymerase”) based on a unique enzyme derived from a bacteriophage infecting Enterococcus faecalis. This polymerase exhibits exceptionally high activity and processivity, including at low and sub-zero temperatures, which distinguishes it from currently available enzymes used in molecular diagnostics.

The project focuses on engineering and optimizing this enzyme for isothermal DNA amplification methods, with the goal of creating a robust, fast and low-temperature diagnostic platform. Such an approach has the potential to significantly improve molecular diagnostics, particularly in point-of-care applications and settings with limited access to advanced laboratory infrastructure. The results of this work will benefit the biotechnology sector, diagnostic companies, clinicians and the scientific community.

The project is carried out in collaboration with Washington University in St. Louis (scientific partner) and QIAGEN (industrial partner).
